The new SNPtower for rapid SNP routine diagnostic is based on a new patent pending method. It combines the advantages of exceptionally fast rapidPCR and simple endpoint detection by using a new method. It makes genotyping of allele conditions extremely simple and fast (for detection of SNP‘s or of point mutations).

 

Sample vessels do not have to be opened after the execution of the assays any longer whereby the risk of contamination is significantly restricted. It so represents an excellent and costeffective alternative to other expensive SNP detection systems.

Measuring method

The analysis is based on the combination of rapidPCR and subsequent endpoint fluorescence detection. The test principle is building on robust homogeneous exonuclease assay. After execution of the exonuclease assays the detection of the fluorescence signals takes place in form of an endpoint determination (not in real-time).
Basis for the final analysis of the allele conditions is the integrated genotyping software ASpect SNP. With this software the analytic evaluation of measured endpoint fluorescence of both probes takes place by means of a multi-component analysis. This analysis makes the accurate determination of the available allele condition possible. This new kind of genotyping is extreme fast and permits the measurement of up to 96 samples in parallel within less than 60 minutes.

 

Prepared for the future

To fit different demands of applications the SNPtower can be equipped with up to 6 excitation and emission filters of choice. These filters are easily interchangeable by the user what keeps the system open for future adaptations or changes. Therefore the instrument is ideally suited for multiplex applications and covers most currently available dyestuffs.

 

Fast detection of PCR products

In addition to SNP diagnostics the SNPtower can be also used as fluorescence reader for easy determination of PCR products. The risk of contamination is significantly restricted due to determination of PCR products without opening the reaction vessel. Particularly for diagnostic applications, simple yes/ no statements are frequently sufficient, e.g. in demonstrating the simple presence of a microorganism in a clinical sample as evidence of an infection.

NOTE: the SNPtower is not applicable for real-time PCR but only for endpoint detection by means of detection before and after the PCR run.

 

 

 

SpeedScan is a fluorescence reader specially designed for easy determination of PCR products. After successful amplificationof the target, direct qualitative evaluation of the PCR can take place by means of fluorescence detection without opening the reaction vessel. The risk of contamination is therefore significantly restricted.

Fast fluorescence detection of PCR products

The polymerase chain reaction (PCR) is one of the absolutely key technologies in modern biological research and molecular diagnostics. Its absence in any biological laboratory is unimaginable. The demands placed on apparatus in the PCR laboratory to guarantee reliable analyses at all times are on the increase, along with the number of PCR applications in routine laboratory work. The type of detection therefore has a crucial influence on the reliability and interpretation of PCR results. The agarose gel used in electrophoresis detection of PCR products is, for example, especially prone to the latent risk of contamination of samples and the laboratory and consequently false-positive results.

 

High speeds and flexibility

PCR end-point detection offers further essential advantages.The introduction of rapid thermal cyclers, such as the SpeedCycler, allows typical amplifications to be performed in down to 8 minutes. Traditional gel electrophoresis with a running time of 30 — 45 minutes plus preparation therefore represents a time restraint. However, the read-out of a 96 well PCR microplate with SpeedScan is completed after 50 seconds per color channel. SpeedScan can record up to 4 different fluorescence filters, which makes it ideally suited for multiplex applications. Naturally  the fluorescence filter is free to use and exchange.

Adapter plate for the SpeedScan validation. The calibration plate contains longterm-stable fluorescence standards, which are based on  special glas. Therefore the software ASpect FA provides a validation tool to calibrate the device by using the calibration plate.
